PENGARUH KRIM EKSTRAK UBI UNGU (Ipomoea Batatas Var Ayumurasaki) TERHADAP MATRIKS METALLOPROTEINASE-1 DAN JUMLAH KOLAGEN DERMIS TIPE-I DAN TIPE-III (Penelitian Eksperimental Pada Mencit BALB/c yang Dipapar Sinar UV-B)

ABSTRAK

 

Latar Belakang : Ubi ungu (Ipomoea Batatas Var Ayumurasaki) terbukti sebagai antiaging karena kandungan antosianin yang bersifat sebagai antioksidan. Antosianin termasuk antioksidan kuat yang dapat melindungi kulit dari radikal bebas. Penelitian mengenai mekanisme kerja ubi ungu ini masih terbatas.

Tujuan : untuk mengetahui pengaruh krim ekstrak ubi ungu terhadap ekspresi MMP-1 dan Jumlah Kolagen Tipe-I dan Tipe-III.

Metode : Penelitian eksperimental menggunakan post test randomized control group design. Dilakukan pada 24 ekor mencit BALB/c yang dibagi 4 kelompok uji. Semua kelompok dipapar dengan sinar UV-B dengan dosis 1 MED (8 menit). Kelompok Kontrol (K) diolesi dengan basis krim, sedangan tiga kelompok perlakuan lainnya diolesi dengan krim ekstrak ubi ungu dengan dosis 0,4%(P1), 4%(P2), dan 40%(P3) segera setelah paparan UV-B yang dilakukan selama 4 minggu. Ekspresi MMP-1 dihitung dari jumlah total fibroblast yang mengekspresikan MMP-1. Ekspresi MMP-1 dan Jumlah Kolagen Tipe-I dan Tipe-III dilakukan dengan pengecatan imunohistokimia. Ekspresi MMP-1 dianalisis dengan One Way Anova dan Post hoc pada tingkat kemaknaan p<0,05

Hasil : Ekspresi MMP-1 antara kelompok K, P1, P2, dan P3 berturut-turut adalah (40,16±4,62), (35,83±3,06), (28,16±2,78), (18,50±2,42) memiliki perbedaan signifikan (p<0,05). Jumlah Kolagen Tipe-I antara kelompok K, P1, P2, dan P3 berturut-turut adalah (48,67±4,58), (55,50±4,80), (64,33±3,55), (67,50±4,18) memiliki perbedaan signifikan (p<0,05). Jumlah Kolagen Tipe-III antara kelompok K, P1, P2, dan P3 berturut-turut adalah (11,83±1,32), (16,16±0,98), (19,67±1,36), (22,00±1,09) memiliki perbedaan yang signifikan antar kelompoknya (p<0,05).

Kesimpulan : krim ekstrak ubi ungu berpengaruh terhadap ekspresi MMP-1 dan Jumlah Kolagen Tipe-I dan Tipe-III pada mencit BALB/c yang dipapar sinar UV-B.

ABSTRACT

Purple sweet potato (Ipomoea Batatas Var Ayumurasaki) is proven to be antiaging because of it is anthocyanin content which is an antioxidant. The antioxidant effects of anthocyanins include powerful antioxidants that protect the skin from free radicals. The mechanism of purple sweet potato's work on the expression MatrixMetalloproteinase-1 (MMP-1) and the Number of Type-I and Type-III Collagen is still limited. The purpose of this study was to determine the purple sweet potato extract cream on expression of MMP-1 and Number of Type-I and Type-III Collagen.

Experimental research using randomized control group design post test. Performed in 24 mice BALB/c were divided four groups test. All groups were exposed to UV-B rays at a dose of 1 MED (8 minutes). The Control Group (K) spread with a base cream, while the other three groups were smeared with cream of purple sweet potato extract at a dose of 0.4% (P1), 4% (P2), and 40% (P3) immediately after exposure to UV-B for four weeks. Expression of MMP-1 was calculated from the total from fibroblasts cells that express MMP-1. Expression of MMP-1 and Number of Type-I and Type-III Collagen by immunohistochemical staining. Expression of MMP-1 was analyzed by One Way Annova and Post hoc at a significance level of p <0.05

Expression of MMP-1 between group C, P1, P2, and P3 are (40.16 ± 4.62), (35.83 ± 3.06), (28.16 ± 2.78), (18.50 ± 2.42) had a significant difference (p <0.05). Number of Type-I Collagen between group C, P1, P2, P3 are (48.67 ± 4.58), (55.50 ± 4.80), (64.33 ± 3.55), (67.50 ± 4.18) had a significant difference (p <0.05). Number of Type-III Collagen between groups C, P1, P2, P3 are (11.83 ± 1.32), (16.16 ± 0.98), (19.67 ± 1.36), (22.00 ± 1.09) had a significant difference between groups (p <0.05).

The conclusion of this study is the purple sweet potato extract cream effects on the expression of MMP-1 and the number of Type-I and Type-III Collagen in BALB /c mice were exposed to UV-B rays.
